No, agents do not reject writers who've self-published (through any means/media) just because they've self-published. If you'd like inspiration: Look for story of David Carnoy.

Jane, How can I get listed in other baking cookbooks as a resource, just wrote a book on home baking. What should I do?
It's a slow process of sending your book to other authors and experts in the field -- as well as getting a highly ranked/reviewed title on Amazon (so that it comes up in any searches).
